Effects of Drying Process of Mistletoes on Cytotoxicities against Cultured HL - 60 and Molt - 4 Cells

Abstract
Aqueous European mistletoe (Viscum album L.) preparation has been used in adjuvant cancer therapy with both immunostimulatory and cytostatic/cytotoxic properties. The cytotoxic and immunological properties of the preparation are considered to be clearly linked to lectins. In this study, the effects of drying process of mistletoe on the cytotoxic activities have been investigated. The cytotoxicity was further reduced by air-drying process than by freeze-drying process, and Molt-4 cells were more sensitive than HL-60 cells to both of air- and freeze- dried samples. The hemagglutinating activity (4 HU) of water extract (200 §·/mL) was not changed by freeze-drying process, whereas the activity disappeared with air-drying process. A galactose-specific lectin, referred as VCA-I, was isolated by affinity chromatogaraphy on asialofetuin-Sepharose 4B. The lectin isolated from freeze-dried plant showed same hemagglutinating activity as fresh one, but the activity of lectin was greatly reduced by air-drying process, indicating that the conformation of lectin in mistletoe may be partially changed by air-drying process. The molecular weight`s of both lectins from fresh and air-dried plant determined by SDS-PAGE were 60,000 kDa, composed of two basic subunits with molecular weights of 31 and 34 kDa of A- and B-chains linked by a disulfide bond. Compared with IC_50 values of water extract, both cancer cells were highly sensitive to lectin. Hence, the results indicate that VCA-I would be the main component for exerting anticancer activities of mistletoe preparation. VCA-I (IC_(50)=1.0 ng/mL) shows similar cytotoxic activity to MLI (IC_(50)=0.7 ng/mL) isolated from European mistletoe.
